Primary cultures were loaded with the calcium indicator Fluo-4 and incubated at 37°C during imaging. Fluo-4 fluorescence intensity is increased in the presence of calcium. Hence, the green flashes occur as a result of calcium entering the cytoplasm. Most of the calcium signals seen occur within astrocytes.
